Development of a novel detection technology for drug resistance mutation sites of Mycobacterium tuberculosis using Luminex liquid chip technology

Purpose: To develop a novel detection technology for drug-resistance mutation sites of Mycobacterium tuberculosis (MTB) using Luminex liquid chip.Methods: Using polymerase chain reaction (PCR) amplification and hybridization analysis, MTB infection drug-resistant the first-line second-line anti-MTB drugs were simultaneously identified. A method was applied to analyze wild-type standard strains 33 clinical samples, results compared with Sanger sequencing PCR products.Results: It revealed that sensitivity (100 %) specificity 31 samples satisfactory, all correctly detected. Compared traditional culture-based drug test, increased speed identification TB, reduced clinicians' workload, decreased treatment cost. Among 12.90 % resistant isoniazid (4/31), 35.48 rifampicin (11/31), ofloxacin (p < 0.05). Furthermore, 2 (6.45 both rifampicin, ofloxacin, 1 (3.22 sample (3.22%) three 0.05).Conclusion: Development wide application this will facilitate MTB, thus reducing spread improving prevention MTB.
